It is not absolutely necessary, but if you plan to NOT use your printer for a couple of days, it is good advice to remove your filament and store it in a dry, cool and dark place. It will make your filament last longer.
For the printer itself, it is not really necessary.
I would recommend removing any translucent filament when finished. I find after 24 hours or so it breaks off in the extruder and can be a pain.
I generally remove the filament if it may be a while before the next print, a day or so, and put it in my humidity controlled storage.
If you are going to change filament and it has been in the machine a while then I would heat, then extrude a bit, like loading, then unload. This will help get rid of a blob or hard area that might get left behind when unloading.
