To be honest, Simplify3D is difficult to explain. Many many options, places to store settings. At the moment I have two FFF profiles, from which you can
work.
I tested all settings in numerous parts and materials, but fine tuning for each print is inevitable.
SINGLE EXTRUDER
Not much to explain.
Basic choices in the top drop down menu’s:
* PLA / ABS / PET material
* fast / medium / high quality
* left / right extruder
On top of that you need to choose:
* infill density / type
* skirt / raft
* support
* cooling
* speed (TAB “other”)
PVA SUPPORT
This is a tricky one. I set up cooling down of the other extruder between layers. Left is base material, right extruder is PVA. Sadly the speed is not independently adjustable for the two materials, so you have to print very slow for PVA to adhere.
Also, you might need to change EXTRUSION MULTIPLIER for PVA much much higher to print well.

* infill density / type
* raft yes / no
* cooling
* tool change script REMEMBER TO CHANGE THE LEFT EXTRUDER TEMPERATURE WHEN CHANGING MATERIAL

DUAL EXTRUSION
Even more tricky to set up, as you need to:
* use the dual extrusion wizard
* have two FFF profiles
* assign the proper FFF profiles to the parts
In the future I hope to elaborate on this subject.
